SUMMARY: Lysis of Mycoplasma gallisepticum by various chemical reagents and enzymes was examined. Lysis was measured by determining the release of intracellular constituents, and compared with the results obtained by the turbidimetric method. The ultracentrifuge was used to compare solutions of isolated membrane in 2-chlorethanol, phenol + acetic acid + water mixture, aqueous urea, and aqueous sodium lauryl sulphate, all of which readily dissolved membranes and whole organisms.
