The Regulation of Phosphoenolpyruvate Carboxykinase and the NADP-linked Malic Enzyme in Aspergillus nidulans
More LessIt has previously been suggested that the synthesis of phosphoenolpyruvate carboxykinase (EC 4.1.1.32) in Aspergillus nidulans is regulated by a repression-derepression mechanism involving a glycolytic intermediate, and not by induction. Results obtained using compounds that enter the tricarboxylic acid cycle via 2-oxoglutarate, and that can supply both a carbon and a nitrogen source for A. nidulans, suggest it is more likely that the synthesis of phosphoenolpyruvate carboxykinase is inducible, and only weakly regulated by carbon catabolite repression. A similar study of the regulation of the NADP-linked malic enzyme (EC 1.1.1.40) indicates that it too may be inducible.

Natural Inhibitors of Fungal Polygalacturonases in Infected Fruit Tissues
More LessIsoelectric focusing of extracts of plum and peach tissue infected with Monilinia spp. revealed the presence of polygalacturonase even though little or no activity was detectable in crude extracts prior to fractionation. Mixing experiments confirmed the presence of an inhibitor of polygalacturonase which was separable from the enzyme by isoelectric focusing. The inhibitor was macromolecular, polydisperse and heat-labile, and its activity was destroyed by trypsin. Similar inhibitors. were found in other host-pathogen combinations.

Sringent Control in an Extreme Thermophilic Bacterium. Preliminary Characterization of a Guanosine 3′,5′-Polyphosphate [(p)ppGpp] Synthesizing Enzyme from Thermus thermophilus
More LessA guanosine 3′,5′-bis(diphosphate) (ppGpp) and guanosine 3′-diphosphate 5′-triphosphate (pppGpp) synthesizing enzyme is present in the ribosomal fraction of Thermus thermophilus. The enzyme is comparable to the Escherichia coli (p)ppGpp synthetase I (stringent factor) in that it requires ribosomes, mRNA and codon-specific, uncharged tRNA. The ribosomal complex can be replaced by methanol. Ribosomes from Escherichia coli do not function as effectors of the Thermus thermophilus enzyme.

DNA:DNA Hybridization Studies of Rhizobium japonicum and Related Rhizobiaceae
More LessThe symbiotic nitrogen-fixing bacteria associated with soybeans (Glycine max) have been classified routinely as Rhizobium japonicum. DNA:DNA hybridization studies were conducted to estimate the genetic relationships of strains labelled as R. japonicum to clarify their taxonomic status. Reassociation reactions were carried out using DNA from various Rhizobium and Agrobacterium species. Relationships of DNA sequences were also estimated by comparing the thermal stability of heterologous DNA duplexes with that of the homologous DNA duplex. The results indicated that strains labelled as R. japonicum can be separated into at least three DNA homology groups. Reference strains of other Rhizobium species, with the exception of R. lupini, were not closely related to these homology groups.
