When incubated at 45° there was no detectable breakdown of DNA in the mutant and the synthesis of deoxynucleoside triphosphates was normal. The temperature-sensitive element affects DNA synthesis in vivo but not in vitro and (unlike an initiator) it appears to be needed for DNA synthesis throughout the replication cycle., UL https://www.microbiologyresearch.org/content/journal/micro/10.1099/00221287-56-1-87
