Infection of cowpea protoplasts with alfalfa mosaic virus RNA was achieved by sedimenting protoplasts from 0·5 m-mannitol and resuspending them in 0·7 m-mannitol containing 0·1 m-potassium phosphate, pH 6·0, and 100 μg/ml RNA. The osmotic shift-up employed during inoculation substantially enhanced the infection by virus RNA as well as by virus nucleoprotein.
As with intact plants, a mixture of the virus genomic RNA species (RNA 1, 2 and 3) was infectious to protoplasts only when supplemented either with virus coat protein or with the messenger RNA for coat protein (RNA 4).
