Abstract

SUMMARY: The effectiveness of a number of different materials in the negative staning method for electron microscopy of viruses is evaluated. Shadowing was used to study the degree of distortion suffered by the specimen. Changes in pH value which occurred while the negative staining solution was trying were measured in those materials which gave the most satisfactory results in the electron microscope. The ultimate resolution of the method is discussed and demonstrated. It was important to de-grease supporting film: perforated carbon films were found valuable for obtaining good contrast.
